EXTRA NOTES

A benzazepine derived from norbelladine. It is found in GALANTHUS and other AMARYLLIDACEAE. It is a cholinesterase inhibitor that has been used to reverse the muscular effects of GALLAMINE TRIETHIODIDE and TUBOCURARINE and has been studied as a treatment for ALZHEIMER DISEASE and other central nervous system disorders.
Cholinesterase inhibitor; reverses scopolamine-induced amnesia; antimyasthenic.

Galantamine hydrobromide (marketed as Reminyl) is one of a group of three drugs for people with Alzheimer's disease called cholinesterase inhibitors. The other two are called donepezil hydrochloride (common brand name Aricept) and rivastigmine (common brand name Exelon). For the purposes of this information sheet, galantamine hydrobromide will be referred to as Reminyl, since it is the brand most people will be familiar with, although other brands may become available. Reminyl is the name given to the particular brand of galantamine hydrobromide produced by a company called Shire Pharmaceuticals. It is licensed for the treatment of mild to moderately severe Alzheimer's disease. Reminyl is not claimed to be a cure for Alzheimer's disease. It treats the symptoms only and there is no evidence that it could halt or reverse the process of brain cell damage that causes Alzheimer's disease.

Local:
Galanthamine (also spelled in galantamine) is a tertiary alkaloid derived from snowdrop and closely related species. It acts as a reversible competitive acetylcholinesterase (AChE) inhibitor, while acts weaker on butyrylcholinesterase (BuChE). It is used in the treatment of disorders of the central nervous system and may be used as an antidote to nonpolarizing muscle relaxants. Galantamine hydrobromide is a white to almost white powder; sparingly soluble in water; insoluble chloroform, ether and alcohol. The chemcial designation is (4aS,6R,8aS)-4a,5,9,10,11,12-hexahydro-3- methoxy-11- methyl-6H-benzofuro[3a,3,2ef]- [2]-benzazepin-6-ol hydrobromide.

GENERAL DESCRIPTION OF ANTICHOLINESTERASE

Anticholinesterase agents (cholinesterase inhibitors) are compounds that prevents the hydrolysis of acetylcholine by acetylcholinesterase and permit high levels of acetylcholine to accumulate at reactive sites.  Reversible competitive or noncompetitive inhibitor of cholinesterase is used in therapeutic uses and as a drug to treat diseases including myasthenia gravis, Alzheimer's disease, and other central nervous system disorders. It is used as an antidote to anticholinergic poisoning. Quasi irreversible inhibitor of cholinesterase is used as a insecticide, and chemical weapon or nerve gas that can be deadly to human or animal.
